The two principal constitutional isomers, 1,2,3-triazole and 1,2,4-triazole, combine aromatic stability with a high density of nitrogen atoms, enabling hydrogen bonding, coordination, dipolar interactions, and diverse substitution patterns. This narrative review provides an integrated introductory account of triazole history, classification, nomenclature, tautomerism, electronic structure, major synthetic strategies, and representative applications. Particular emphasis is placed on the transformation of 1,2,3-triazole chemistry by copper-catalysedazide-alkyne cycloaddition and on complementary synthetic routes to 1,2,4-triazoles. The review also distinguishes established clinical uses of triazole-containing medicines from preclinical biological activity reported for experimental derivatives. Peer-reviewed literature was identified through searches of PubMed and publisher-hosted journal databases through September 2026, with priority given to landmark primary studies, authoritative reviews, and records with verifiable bibliographic metadata and digital object identifiers. Beyond medicinal chemistry, triazoles are relevant to bioconjugation, polymer and materials science, corrosion inhibition, and agrochemical research. Current opportunities include greener and more selective synthesis, improved control of regio- and stereochemistry, mechanistically informed medicinal chemistry, and more rigorous translation of screening activity into pharmacologically validated leads. Together, these features explain the continuing value of triazoles as versatile scaffolds across chemical research.
Karan Singh Nehal Sagwal Sunil Kumar Copyright (c) 2026 Author(s). Their physicochemical stability, capacity for hydrogen bonding and dipolar interactions, and compatibility with diverse substitution patterns have made triazoles important scaffolds in synthetic and medicinal chemistry. This narrative review summarises established and recent strategies for the synthesis of 1,2,3- and 1,2,4-triazoles, approaches used to distinguish regioisomers, and representative biological applications of triazole-containing compounds. Particular attention is given to thermal and metal-catalysed azide-alkyne cycloadditions, metal-free approaches, 1,2,4-triazole-forming reactions, anticancer, anti-inflammatory, antimicrobial, and antiviral activities, selected clinically used and investigational triazole-containing compounds, toxicity considerations, and emerging applications. The evidence is interpreted according to the level of experimental support, with computational, biochemical, cellular, animal, and clinical findings distinguished where relevant.
Hassan Gamal Khadiga A. This narrative review summarises their principal structural classes, representative synthetic strategies, and selected biological applications, with emphasis on 1,2,4-triazolopyrimidine systems. Major routes include annulation of pyrimidine rings onto aminotriazoles, annulation of triazole rings onto functionalised pyrimidines, cyclocondensation, multicomponent reactions, oxidative ring closure, and greener solvent-free or bio-based catalytic methods. The scaffold also serves as a precursor to higher fused and polycyclic heterocycles. Source-verified studies illustrate antiproliferative, antimicrobial, antiviral, antimalarial, anticonvulsant, antioxidant, enzyme-inhibitory, and corrosion-inhibition applications. Structure-activity studies show that substitution pattern and regiochemistry can markedly alter potency and target engagement; however, cross-study comparisons are constrained by heterogeneous assays and limited translational validation. Recent work has broadened the pharmacological scope to selective ENPP1 inhibition, multitarget anticancer hybrids, alpha-glucosidase inhibition, and greener antimicrobial synthesis. Overall, triazolopyrimidines remain versatile frameworks for synthetic diversification and early-stage lead discovery, while further development requires rigorous evaluation of selectivity, pharmacokinetics, toxicity, reproducibility, and in vivo performance.
Raman Lakhia Sidhant Yadav Poonam Devi Karmvati Yadav Rashmi Pundeer Copyright (c) 2026 Author(s). This narrative review examines representative strategies for the synthesis of [1,2,4]triazolo[3,4-b][1,3,4]thiadiazole derivatives and critically summarises structure–activity relationships across anticancer, antimicrobial, acetylcholinesterase-inhibitory, anti-inflammatory, analgesic, anticonvulsant, and emerging urease-inhibitory applications. The literature was updated through 27 September 2026 by targeted searches of PubMed, journal and publisher platforms, and DOI-based bibliographic records, with peer-reviewed primary studies prioritised for quantitative biological data. Common synthetic routes proceed through 4-amino-5-substituted-4H-1,2,4-triazole-3-thiols followed by cyclocondensation with carboxylic acids, aldehydes, or related electrophiles; microwave-assisted and visible-light-mediated variants have also been reported. Across individual series, halogen, alkoxy, nitro, aryl, glycosyl, and lipophilic substituents can alter activity, but these effects are assay- and scaffold-dependent rather than universally predictive. Representative studies have reported micromolar antiproliferative and acetylcholinesterase-inhibitory activity, antimicrobial effects at low microgram-per-milliliter concentrations, preclinical anti-inflammatory and anticonvulsant activity, and submicromolar-to-low-micromolar urease inhibition. Most evidence remains preclinical and heterogeneous, with limited pharmacokinetic, selectivity, and repeated-dose toxicity data. The scaffold therefore remains a useful platform for medicinal and bioactive chemistry, while standardised comparative assays, target validation, and in vivo translational studies are required to establish therapeutic relevance.
Ola Mohamed Nesreen Saied Haiba Mohamed Teleb Hayam M.A. Ashour Sherry N. This narrative review critically examines thiazolo[3,2-b][1,2,4]triazoles, thiazolo[2,3-c][1,2,4]triazoles, and the much less well represented isothiazolo[3,2-c][1,2,4]triazole framework. The synthetic literature is evaluated by reaction logic rather than chronology alone, with emphasis on sulfur alkylation followed by cyclisation, Hantzsch-type condensations, dehydrative cyclisation, Sonogashira-enabled annulation, and visible-light-mediated aqueous synthesis. Particular attention is given to regioisomer assignment, structural proof, substrate scope, operational limitations, and sustainability. The biological section prioritises primary preclinical studies and distinguishes biochemical, cell-based, and animal evidence. Representative data include antibacterial and antifungal minimum inhibitory concentrations, antiproliferative GI50 values with p53-associated responses, anticonvulsant ED50 and TD50 values in mouse seizure models, and enzyme-inhibition studies involving carbonic anhydrase, urease, cyclooxygenase, and butyrylcholinesterase. In relation to previous broad reviews, the present article emphasises route-to-regioisomer relationships, explicit scaffold boundaries, primary-evidence appraisal, and recent synthetic and biological developments through 2026. The available evidence supports continued lead-discovery and synthetic-methodology research, but it remains predominantly preclinical; rigorous selectivity, toxicity, pharmacokinetic, and reproducibility studies are required before therapeutic significance can be established.
Anita Anita Karan Singh Nehal Sagwal Sunil Kumar Dinesh Kumar Copyright (c) 2026 Author(s).
