Abstract
Thiazolotriazoles are fused sulfur- and nitrogen-containing heterocycles that combine a 1,2,4-triazole nucleus with a thiazole or isothiazole ring and provide structurally diverse platforms for heterocyclic and medicinal chemistry. This narrative review critically examines thiazolo[3,2-b][1,2,4]triazoles, thiazolo[2,3-c][1,2,4]triazoles, and the much less well represented isothiazolo[3,2-c][1,2,4]triazole framework. The synthetic literature is evaluated by reaction logic rather than chronology alone, with emphasis on sulfur alkylation followed by cyclisation, Hantzsch-type condensations, dehydrative cyclisation, Sonogashira-enabled annulation, and visible-light-mediated aqueous synthesis. Particular attention is given to regioisomer assignment, structural proof, substrate scope, operational limitations, and sustainability. The biological section prioritises primary preclinical studies and distinguishes biochemical, cell-based, and animal evidence. Representative data include antibacterial and antifungal minimum inhibitory concentrations, antiproliferative GI50 values with p53-associated responses, anticonvulsant ED50 and TD50 values in mouse seizure models, and enzyme-inhibition studies involving carbonic anhydrase, urease, cyclooxygenase, and butyrylcholinesterase. In relation to previous broad reviews, the present article emphasises route-to-regioisomer relationships, explicit scaffold boundaries, primary-evidence appraisal, and recent synthetic and biological developments through 2026. The available evidence supports continued lead-discovery and synthetic-methodology research, but it remains predominantly preclinical; rigorous selectivity, toxicity, pharmacokinetic, and reproducibility studies are required before therapeutic significance can be established.
