Abstract
Triazolopyrimidines are fused nitrogen-containing heterocycles with broad utility in synthetic and medicinal chemistry. This narrative review summarises their principal structural classes, representative synthetic strategies, and selected biological applications, with emphasis on 1,2,4-triazolopyrimidine systems. Major routes include annulation of pyrimidine rings onto aminotriazoles, annulation of triazole rings onto functionalised pyrimidines, cyclocondensation, multicomponent reactions, oxidative ring closure, and greener solvent-free or bio-based catalytic methods. The scaffold also serves as a precursor to higher fused and polycyclic heterocycles. Source-verified studies illustrate antiproliferative, antimicrobial, antiviral, antimalarial, anticonvulsant, antioxidant, enzyme-inhibitory, and corrosion-inhibition applications. Structure-activity studies show that substitution pattern and regiochemistry can markedly alter potency and target engagement; however, cross-study comparisons are constrained by heterogeneous assays and limited translational validation. Recent work has broadened the pharmacological scope to selective ENPP1 inhibition, multitarget anticancer hybrids, alpha-glucosidase inhibition, and greener antimicrobial synthesis. Overall, triazolopyrimidines remain versatile frameworks for synthetic diversification and early-stage lead discovery, while further development requires rigorous evaluation of selectivity, pharmacokinetics, toxicity, reproducibility, and in vivo performance.
